Most turnbuckles have a hole drilled in the end of the threaded portion so when it come through the turn buckle body you put a cotter ring or cotter pin through it so it can't unscrew itself. If there isn't a hole then on of the threaded portions of the turn buckle screw you should have a nut on it so it can be tightened against the turnbuckle body to lock it in place. When it comes to how far the threaded portion needs to into the body? It depends as to what the body is made of. If you look at any nut about that size they have 4 or 5 threads in them and they work. So you need to judge how hard the material is, what condition the thread are in and if it's just 45 or 5 threads you will need to use a lock nut to not let the threads "work".
